By day, Ryu Hayabusa is the owner of a simple shop selling Japanese antiques. By night, he defends New York City as the Dragon Ninja. When the respected Dr. Ned Friedman announces that he has developed a perfect cure for cancer in an extremely bizarre press conference, Ryu knows that something is very wrong. Along with his trusted associates who work for a small newspaper, Ryu sets out to uncover the dark secrets behind Dr. Friedman's supposed miracle cure.

Veteran adventurer Nick, mage Tiana, cleric Zem, and half-dragon Curran each show up alone at a tavern, only to find themselves seated at the same table. As the night goes on, they take turns divulging their grievances and the hobbies they have taken up as a means of coping. While their stories are utterly different, there is one common thread: betrayal. Each of the four, having been bitterly betrayed by someone they treasured, has developed a deep-seated distrust of humanity. But in addition to their similar worldviews, the four have one more commonality: a critical lack of funds. Realizing that things cannot continue as they are, they decide to form a party, rank up, and make as much money as possible to spend on their respective hobbies. What these disillusioned adventurers do not know, however, is that they will one day save the world from unfathomable peril.

Nichijou primarily focuses on the daily antics of a trio of childhood friends—high school girls Mio Naganohara, Yuuko Aioi and Mai Minakami—whose stories soon intertwine with the young genius Hakase Shinonome, her robot caretaker Nano, and their talking cat Sakamoto. With every passing day, the lives of these six, as well as of the many people around them, experience both the calms of normal life and the insanity of the absurd. Walking to school, being bitten by a talking crow, spending time with friends, and watching the principal suplex a deer: they are all in a day's work in the extraordinary everyday lives of those in Nichijou.
